The pathogen isolation, identification and Physiological properties of peanut crown rot in Liaoning Province
The purpose of this study was to identify the pathogens of peanut crown rot in Liaoning Province. The pathogens were isolated by tissue isolation method and verified by Koch 's rule. Then the colony
mycelium and spore morphology of the pathogen were observed. The results showed that the strain exhibited circular colony growth with neat edges
white fluffy mycelia featuring septa and branching
and black spherical conidia. Based on multigene phylogenetic analysis of
ITS
CAL
and
Tub2
partial sequences
the pathogen causing peanut crown rot in Liaoning Province was identified as “
Aspergillus niger
”. In addition
the Physiological properties of the pathogen were tested. The results showed that the optimum growth temperature of the pathogen was 35°C
the optimum growth pH was 5
the optimum growth carbon and nitrogen sources were lactose and tryptone
