Establishment of obese mouse models by simulating different dietary habits and comparison of their intervention outcomes
To establish obese mouse models by simulating different dietary habits
and compare their mechanisms and intervention effects.
Methods
2
A total of 42 5‐week‐old male C57BL/6 mice were randomly divided into a high‐sugar‐high‐fat group (HSHF)
a simple high‐fat group (HF)
and a blank control group (Control)
and fed with corresponding feed for 12 weeks. Body mass
body length
body width
and blood glucose were monitored every week
and the success of the obesity model was determined by a Lee index ≥340 for two consecutive weeks. After modeling
7 animals were euthanized in each group to observe intra‐abdominal fat accumulation and liver oil red O staining (to evaluate lipid droplet deposition). The remaining mice were switched to regular maintenance feed and regular exercise for 2 hours daily. After 4 weeks of intervention
they were euthanized and assessed for fat accumulation.
Results
2
During the modeling period (0‐12 weeks)
the indicators of each group increased over time
and the differences between the experimental groups and the control group gradually widened. At week 12
the body weight of the HSHF group was significantly higher than that of the HF group ([46.98±1.83
]
g versus [43.55±1.41
]
g
P
<
0.01) and the Control group (versus [36.42±1.12
]
g
P
<
0.000 1). The Lee indexes of both the HSHF and HF groups remained above 340 for two consecutive weeks and were significantly higher than those of the Control group
indicating the successful establishment of the obesity models. The blood glucose levels in the experimental groups were also significantly higher than those in the control group. Liver oil red O staining staining showed significantly increased and extensively distributed lipid droplet deposition in the experimental groups. During the intervention period (13‐16 weeks)
the body mass
Lee index
and blood glucose of all three groups showed a decreasing trend.
Conclusion
2
Two mouse models of obesity were successfully established. Long⁃term feeding of a high⁃glucose⁃high⁃fat diet significantly increased body weight
Lee index
blood glucose
and fat accumulation
confirming the link between excessive caloric intake and obesity/metabolic disorders. The observed hepatic pathological changes revealed the mechanism underlying visceral injury. Dietary modification combined with regular exercise effectively improved obesity and related metabolic parameters
providing an evidence⁃based intervention strategy for prevention and treatment.
