摘要

目的

通过孟德尔随机化分析来探究吸烟、饮酒和喝咖啡与骨关节炎之间的因果关联。

方法

从已发表的全基因组关联研究（GWAS）中选择并提取吸烟、饮酒和喝咖啡这3个生活习惯相关特征的工具变量，手骨关节炎、膝骨关节炎和髋骨关节炎的工具变量也来自公开的全基因组关联研究。采用逆方差加权法（IVW）作为主要分析方法，同时采用孟德尔随机化⁃Egger回归、加权中位数法、MR⁃PRESSO等方法检测和校正结果。

结果

逆方差加权法结果表明，吸烟与骨关节炎之间存在因果关联（膝骨关节炎：

0.05，膝和/或髋骨关节炎：

0.05），喝咖啡与骨关节炎之间存在因果关联（膝骨关节炎：

=0.01，膝和/或髋骨关节炎：

=0.03），饮酒与骨关节炎之间没有因果关联（

0.05）。

结论

吸烟和喝咖啡与骨关节炎之间存在因果关联，而没有足够的证据表明饮酒和骨关节炎之间存在因果关联。

Abstract

Objective

To investigate the association between smoking

drinking

coffee consumption

and the risk of osteoarthritis through Mendelian randomization analysis.

Methods

Instrumental variables for smoking

alcohol consumption

and coffee consumption were selected and extracted from published genome⁃wide association studies (GWAS)

and instrumental variables for hand osteoarthritis

knee osteoarthritis

and hip osteoarthritis were also derived from publicly available genome⁃wide association studies. The inverse variance weighting (IVW) method was used as the main analysis method

and MR⁃Egger regression

weighted median method

MR⁃PRESSO

and other methods were used to detect and correct the results.

Results

The results of the inverse variance weighting method showed that smoking was positively causal with the risk of osteoarthritis (knee osteoarthritis:

0.05; knee and/or hip osteoarthritis:

0.05)

coffee consumption was positively causal with the risk of osteoarthritis (knee osteoarthritis:

=0.01; knee and/or hip osteoarthritis:

=0.03)

and there was no causal association between drinking and osteoarthritis (

0.05).

Conclusion

There is a genetic causal association between smoking and coffee consumption and osteoarthritis

whereas there is insufficient evidence for a genetic causal association between drinking and osteoarthritis.
