Identification of psoriasis signature genes and key pathways based on bioinformatics methods
To identify the psoriasis characteristic genes and key pathways for a deeper understanding of the pathogenesis of psoriasis by using bioinformatics methods.
Methods
2
GSE6710 data was obtained from gene expression omnibus (GEO)
and different expression analysis was performed for the corrected chip expression profile by using the limma package in R. GO/KEGG analysis was performed to identify the biological processes and pathways involved by the differentially expressed genes
subsequently. These genes were imported into the STRING database for protein interaction analysis. Topological analysis of the protein interaction network was performed by the CytoNCA plugin to identify signature genes and perform functional annotation.
Results
2
A total of 90 differentially expressed genes (64 up⁃regulated genes
26 down⁃regulated genes) and 6 signature genes (CXCL2
CXCL1
CCL4
CCL20
CXCL8
and LCN2) were selected. These genes were highly expressed in psoriatic lesion tissues
mainly involved in chemokines⁃mediated signaling pathway
viral protein and cytokine and cytokine receptor interaction
IL⁃17 signaling pathway
neutral leukocyte migration
and other biological processes and pathways.
Conclusion
2
CXCL2
CXCL1
CCL4
CCL20
CXCL8
and LCN2 were the key genes related to the occurrence and development of psoriasis
which were mainly involved in the inflammatory response process of the body.
